What is PNC-27, and how is it thought to work?
p53 is a protein known as a “tumor suppressor” that regulates the cell’s response to DNA damage. HDM-2 (MDM2 in mice) is a protein that binds p53 and suppresses it. PNC-27 is a synthetic 32-amino-acid peptide that links amino acids 12–26 of p53, corresponding to the portion that binds HDM-2, to a carrier sequence (penetratin) that allows it to cross the cell membrane. [3]
According to a study published in 2010 by the group that developed the peptide, PNC-27 kills cancer cells by a route different from conventional drugs, by breaking apart the cell membrane (membranolysis). The researchers reported finding substantial levels of HDM-2 in the membrane of various cancer cells but not in the membrane of several non-cancerous cell lines tested. When HDM-2 was placed in the membrane of non-cancerous breast epithelial cells, these cells also became sensitive to PNC-27. From this the authors conclude that the selectivity of the peptide stems from HDM-2 in the membrane. [2]
In a 2022 study the same group reported seeing, by electron microscopy, ring-shaped pores in the cancer cell membrane in which PNC-27 and HDM-2 were located together, and no pore formation in non-cancerous fibroblasts treated with the same peptide. [3] This mechanism has never been shown in humans.
What do the laboratory studies show?
Most of the published data on PNC-27 are cell culture studies. In these studies the peptide killed cancer cell lines carrying HDM-2 in their membrane and had no effect on the normal cell lines tested. [2] [3]
Three limits matter when reading these findings:
- Narrow range of sources: Most publications come from the research group that developed the peptide. According to the conflict-of-interest statement of the 2022 paper, three of the authors are inventors of intellectual property and patents related to PNC-27 and PNC-28; the institution of the first author is a pharmaceutical company. [3]
- Cell culture ≠ body: A peptide killing cells in a laboratory dish does not show that it will reach a tumor when given into the blood, that enough of it will remain without being broken down, or that it will not harm healthy tissues.
- “Normal cell” is limited: Selectivity was tested in a limited number of non-cancerous cell lines and in a culture environment. [2]
What was found in animal experiments?
| Molecule | Model | Reported finding | Limit | Grade |
|---|---|---|---|---|
| PNC-28 (amino acids 17–26 of p53 + penetratin; shorter analog of PNC-27) | Ras-transformed rat pancreatic cancer cells transplanted into nude mice | When given into the abdomen for 2 weeks at the same time as the tumor, the tumors disappeared completely; growth of the tumor at a distant site stopped or slowed compared with the control peptide [4] | Not a human tumor; not PNC-27; only the developing group | E — not studied in humans |
| PNC-27 | Human and mouse acute myeloid leukemia (AML) models, primary and secondary transplantation experiments | Killed leukemia cells and subgroups rich in leukemia stem cells; preserved normal blood-forming stem cell function in wild-type mice [5] | In mice; a company supplied the peptide for the study | E — not studied in humans |
The 2020 leukemia study was carried out by a team from centers outside the group that developed the peptide, mostly in the US and China; however, according to the conflict-of-interest statement, a company supplied PNC-27 and the control peptides. [5] The authors of the 2006 pancreatic study also phrase the result as a possibility: “PNC-28 may be effective in the treatment of cancers, especially if given directly into the tumor.” [4] Mouse results represent an early stage at which candidate cancer drugs can still fail in humans, and they are not a treatment recommendation.
Has PNC-27 been tested in humans?
There is no registered study. A search for “PNC-27” on ClinicalTrials.gov returns no registered clinical trial as of October 19, 2026. [6] In its 2020 and 2021 statements the US Department of Justice states that PNC-27 has not been approved by the FDA as a drug for the treatment of any disease, including cancer, and has not undergone clinical trials in humans in the US. [7] [8]
Despite this, PNC-27 has been offered to cancer patients as an “alternative treatment.” According to the FDA, products could be sold in various forms such as inhaled solution, intravenous solution, and vaginal and rectal suppositories. [1] Because there is no registered human study, neither the effectiveness nor the safety of these uses is known; no safe form of use has been defined.
What did the FDA and US authorities do?
2017 FDA warning: bacteria in products
On January 10, 2017, the FDA warned consumers not to buy or use PNC-27 marketed as a cancer treatment through a website. The agency’s laboratory found the bacterium Variovorax paradoxus in a sample of inhaled PNC-27 solution; an update on March 27, 2017 added that the bacterium Ralstonia insidiosa was found in another sample. The FDA stated that people using contaminated products face a risk of serious and potentially life-threatening infection, and that young children, older adults, pregnant women and people with weakened immune systems are at higher risk. The agency wrote that it has not evaluated or approved PNC-27 as safe and effective for any disease, including cancer. As of the date of the warning, the FDA had received no report of illness or serious adverse event associated with PNC-27. [1]
The FDA’s cancer-fraud question-and-answer page published the same year states that illegally sold cancer products can cause direct harm and can cause indirect harm by delaying proven treatments or interacting with them; as an example of direct risk it cites the bacterial finding in the PNC-27 case. [9]
Criminal case: suppositories made in a kitchen from a peptide bought “for research”
According to the 2020 indictment filed in Alabama, in 2015–2016 a person bought PNC-27 from a China-based manufacturer and repeatedly assured the manufacturer that the peptide would be used only in laboratory research. In reality the peptide was turned into suppositories in a non-sterile kitchen and warehouse that did not comply with good manufacturing practices; some customers reported finding pieces of hair in the suppository packages. The products were marketed to alternative-medicine physicians, cancer patients and an alternative cancer treatment center in Mexico. [7]
In January 2021 the defendant pleaded guilty to conspiracy to fraudulently introduce adulterated and misbranded drugs into commerce. In May 2021 the court imposed a 30-month prison sentence followed by 3 years of supervised release and forfeiture of $900,000. According to the prosecutor’s statement, the defendant had presented the distribution as a “research study” and presented himself as a research director; he had no training in medicine or disease treatment. [8]
This case concretely shows that a “for research use only” label does not mean that a product is safe or legal for human use. The meaning of the label is covered in a separate guide:
Detailed guideWhat does 'research peptide' mean? What do other countries say?
Health Canada, Canada’s health authority, issued a public advisory in April 2025 about unauthorized injectable peptides seized from a wellness center in Calgary; “PNC 27” was on the list. The agency stated that it had not authorized any of the seized products, that they had not been assessed for safety, efficacy and quality, and that their sale in Canada is illegal. According to Health Canada, unauthorized injectable drugs can cause infections, allergic reactions and drug interactions, may contain high-risk ingredients or contaminants not listed on the label, and may not have been manufactured or stored under safe conditions. [10]
Is PNC-27 prohibited in sport?
The WADA 2026 Prohibited List does not name PNC-27. The S0 class of the list prohibits, in and out of competition at all times, pharmacological substances that are not covered in any other section of the list and that no governmental health authority has approved for human therapeutic use (including drugs in preclinical or clinical development). [11] The FDA has not evaluated or approved PNC-27 for any disease; because the S0 definition depends on approval by a governmental health authority, this substance, which has no approval document, is directly relevant for licensed athletes. [1]
What is the status of PNC-27 in Türkiye?
Status in Türkiye
- License
- No product containing PNC-27 appears on the TİTCK (Turkish Medicines and Medical Devices Agency) Licensed Human Medicinal Products List dated October 16, 2026 [12]
- Law
- Law No. 1262 on Pharmaceutical and Medicinal Preparations (İspençiyari ve Tıbbi Müstahzarlar Kanunu); Law No. 6197 on Pharmacists and Pharmacies (Eczacılar ve Eczaneler Hakkında Kanun) [13] [14]
- Sport
- The WADA S0 definition always prohibits unapproved substances [11]
Article 3 of Law No. 1262 requires Ministry permission before preparations are placed on the market and before those produced abroad are imported. Article 19 provides imprisonment of one to five years for those who produce unlicensed preparations and for those who knowingly sell, offer for sale or have them sold; it ties the sale, marketing and advertising of a product to the same penalty when it is claimed to diagnose or treat diseases, even if it is not a preparation, and treats the commercial import of a preparation produced abroad without a license as smuggling. Additional Article 7 provides that the Ministry may permit the import of unlicensed preparations for examination, experiment or personal treatment, provided that the amount does not exceed the quantity to be accepted by the Ministry. [13] Under Article 24 of Law No. 6197, medicines cannot be sold on the internet or any other electronic medium. [14] How products sold with a “cancer treatment” claim or a “for research purposes” label stand under these provisions is assessed in the specific case by the administration and the courts; this page is not legal advice.

What are the risks of PNC-27?
The risk picture for PNC-27 consists of unknowns and product quality more than known side effects:
- Contamination: The FDA found bacteria in two separate PNC-27 samples; contaminated products carry a risk of serious infection. [1]
- Manufacturing conditions: In the US case, the products were prepared in a non-sterile kitchen without adherence to good manufacturing practices. [8]
- Unknown safety profile: Because there is no registered human study, which side effects occur, and how often, is not known. [6]
- Delay of treatment: The FDA stresses that unproven cancer products can cause indirect harm by delaying effective treatments or interacting with them. [9]
- Injectable products: According to Health Canada, unauthorized injectable peptides carry a risk of infection, allergic reaction and undeclared contaminants. [10]
